ABSTRACT

Abstract Piper nigrum (black pepper) is used in Indian traditional medicine and its main alkaloid, Piperine (PIP), presents antioxidant, antitumor and neuroprotective pharmacological properties. This substance is insoluble in aqueous media and can irritate the gastrointestinal tract. Aiming to avoid these inconvenient characteristics and enable PIP oral administration, this study suggested the PIP microencapsulation through the emulsion-solvent evaporation method and the preparation of microparticulated tablets by direct compression. An UV-spectroscopy method was validated to quantify PIP. Microparticles and microparticulated tablets were successfully obtained and the microparticles exhibited excellent flow. The scanning electron microscopy images showed that PIP microparticles were intact after compression. The in vitro release showed a controlled release of PIP from microparticles and PIP microparticles from tablets in comparison to PIP and PIP tablets. The release profiles of PIP microparticles and the microparticulated tablets were similar. Therefore, tablets containing PIP microparticles are promising multiparticulated dosage forms because a tablet allows microparticles administration and the intact ones promote a controlled release, decreasing its irritating potential on the mucosa.

ABSTRACT

ABSTRACT: Technique of complex coacervation was used to produce microcapsules of Lactobacillus acidophilus La-5 encapsulated in gelatin and gum arabic which were then freeze-drying. Microcapsules were characterized using scanning electron and optical microscopy, and resistance of probiotics was evaluated during release into a simulated gastrointestinal tract and storage at different temperatures. The complex coacervation process produced microcapsules with a high encapsulation efficiency (77.60% and 87.53%), ranging from 127.14-227.05 μm with uniform distribution. Microencapsulation was an efficient approach to achieve significant protection of probiotics against simulated gastrointestinal conditions compared with free cells. Encapsulation also improved the viability of probiotics during storage at either −18 ºC for 120 days, 7 ºC for 105 days or 25 ºC for 45 days. Therefore, complex coacervation was demonstrated to be adequate and promising for encapsulation of probiotics.


RESUMO: A técnica de coacervação complexa foi utilizada para a produção de microcápsulas contendo Lactobacillus acidophilus La-5 em gelatina e goma arábica seguida de secagem por liofilização. As microcápsulas foram caracterizadas por microscopia óptica e eletrônica de varredura, assim como a resistência dos probióticos frente à liberação "in vitro" ao trato gastrointestinal simulado e ao armazenamento em diferentes condições de temperatura também foram avaliados. O processo de coacervação complexa formou microcápsulas com alta eficiência de encapsulação (77,60% e 87,53%), tamanho compreendido entre 127,14 e 227,05 µm e distribuição uniforme. As microcápsulas foram eficientes em promover a proteção substancial dos probióticos frente às condições gastrointestinais simuladas, em comparação às células livres. A encapsulação também foi eficiente em manter a viabilidade dos probióticos durante o armazenamento em temperaturas de −18 ºC por 120 dias, 7 ºC por 105 dias e 25 ºC por 45 dias. Dessa forma, a coacervação complexa se mostra adequada e promissora para a encapsulação dos probióticos.

ABSTRACT: This study produced pectin microcapsules containing Lactobacillus acidophilus by external ionic gelation, followed by the adsorption of whey protein and pectin to form multilayers. The viability of free and microencapsulated lactobacilli was evaluated after in vitro exposure to gastrointestinal conditions. They were also assessed by heat treatment, and stability was examined at -18 °C, 5 °C and 25 °C for 120 days. Exposure to different pHs, simulating passage through the gastrointestinal tract, showed that treatment of the microcapsules with only pectin (LA/P0) and with one and two layers of whey protein (treatments LA/P1 and LA/P3, respectively), were able to protect Lactobacillus acidophilus , with microcapsules increasing the release of probiotics from the stomach into the intestines. Free cells showed a decrease in their counts over the course of the simulated gastrointestinal system. Regarding heat treatments, microcapsules with a layer of whey protein (LA/P1) maintained the viability of their encapsulated Lactobacillus acidophilus (9.57 log CFU/g-1). The best storage viability was at -18 °C, with a count of 7.86 log CFU/g-1at 120 days for microcapsule LA/P1,with those consisting of two layers of whey protein (LA/P3)having a 6.55 log CFU/g-1 at 105 days. This study indicated that external ionic gelation was effective and could be used for the production of pectin microcapsules, with multilayer whey protein promoting greater protection and viability of Lactobacillus acidophilus.


RESUMO: O objetivo deste trabalho foi produzir microcápsulas de pectina, contendo Lactobacillus acidophilus por gelificação iônica externa, seguida da adsorção de proteína de soro de leite e multicamadas formadoras de pectina. Além disso, a viabilidade de lactobacilos livres e microencapsulados, após exposição in vitro a condições gastrintestinais, foi avaliada após simulação de tratamentos térmicos e, finalmente, estabilidade a -18 ºC, 5 ºC e 25 ºC durante 120 dias de armazenamento. A exposição a diferentes pHs, simulando a passagem pelo trato gastrointestinal, mostrou que os tratamentos das microcápsulas com apenas pectina (LA/P0) e com uma e duas camadas proteína do soro (tratamentos LA/P1 e LA/P3, respectivamente), foram capazes de proteger o Lactobacillus acidophilus , enquanto as microcápsulas aumentaram a liberação de probióticos do estômago para o intestino. As células livres diminuíram suas contagens no curso do sistema. Em relação aos tratamentos térmicos aplicados, pode-se afirmar que a microcápsula com uma camada de proteína do soro (LA/P1) resistiu e manteve a viabilidade de Lactobacillus acidophilus (9,57 log CFU / g-1). A melhor viabilidade foi obtida no armazenamento a -18 ° C, com uma contagem de 7,86 log CFU / g-1 para essa mesma microcápsula (LA/P1) no final do armazenamento (120 dias) e 6,55 log CFU / g-1 para as microcápsulas com duas camadas de proteína do soro (LA/P3) por 105 dias. Este estudo indica que a gelificação iônica externa é eficaz e pode ser usada para a produção de microcápsulas de pectina com multicamadas de proteína de soro para promover maior proteção e viabilidade ao Lactobacillus acidophilus.

ABSTRACT

ABSTRACT: The high intensity ultrasound-assisted extraction (HIU) is one of the most simple, quick and efficient techniques for the extraction of phenolic and other antioxidant compounds from plants. This is the first application of HIU for the extraction of these compounds from goldenberry fruit. The HIU and conventional extraction techniques showed similar results regarding to phenolic compounds and antioxidant capacity. However, the time required for HIU extraction (5min) was 24 times lower than conventional extraction (120min). Phenolic compounds reported were chlorogenic acid, caffeic acid and rutin. In vitro cytotoxicity assays were used for evaluation of extracts and the results showed that in a wide range of concentration, the extract maintains cell viability, thus indicating the possibility to use it as food with safety.


RESUMO: A extração assistida com ultrassom de alta intensidade (HIU) é uma das técnicas mais simples, rápidas e eficientes na extração de compostos fenólicos e antioxidantes de plantas. Este trabalho foi o primeiro a utilizar HIU na extração destes compostos presentes na fruta goldenberry. As técnicas HIU e extração convencional apresentaram resultados semelhantes com relação aos compostos fenólicos e capacidade antioxidante. Entretanto, o tempo necessário na HIU (5min) foi 24 vezes menor que na extração convencional (120min). Os compostos fenólicos encontrados foram ácido clorogênico, ácido cafeico e rutina. Ensaios de citotoxicidade in vitro foram usados para avaliação dos extratos e os resultados demonstraram que, em ampla faixa de concentração, o extrato mantém a viabilidade celular, indicando assim possível segurança para utilização em alimentos.

ABSTRACT: Lactobacillus acidophillus La-5 (ML) and Bifidobacterium Bb-12 (MB) microparticles were produced at different temperatures by spray dryer. The influence of different temperatures on the viability, encapsulation efficiency, water activity and moisture were evaluated. Microparticles that presented more viability were submitted to thermal resistance, gastrointestinal simulation, storage stability, morphology and particle size analyses. Drying temperature of 130°C showed higher encapsulation efficiency, 84.61 and 79.73% for Lactobacillus acidophillus (ML) and Bifidobacterium Bb-12 (MB) microparticles, respectively. In the evaluation of thermal resistance and gastrointestinal simulation, the microparticles of Lactobacillus acidophillus La-5 (ML) presented higher survival than Bifidobacterium Bb-12 (MB) under these conditions. In storage viability only the Lactobacillus acidophillus La-5 (ML) microparticles remained viable at all evaluated temperatures during the 120 days. The particle sizes reported were 4.85 for Lactobacillus acidophillus La-5 (ML) and 8.75 for Bifidobacterium Bb-12 (MB), being in agreement with the desired values for products obtained by spray dryer. Finally, the Lactobacillus acidophilus La-5 (ML) microparticles were shown to be more resistant under the conditions evaluated in this study.


RESUMO: Micropartículas de Lactobacillus acidophillus La-5 (ML) e Bifidobacterium Bb-12 (MB) foram produzidas em diferentes temperaturas de secagem no spray dryer. A influência das diferentes temperaturas sobre a viabilidade, eficiência de encapsulação, atividade de água e umidade foram avaliadas. As micropartículas que apresentaram maior viabilidade foram submetidas a análises de resistência térmica, simulação gastrointestinal, estabilidade ao armazenamento, morfologia e tamanho de partícula. A temperatura de secagem de 130°C mostrou maior eficiência de encapsulação, 84.61 e 79.73% para micropartículas de Lactobacillus acidophillus (ML) e Bifidobacterium Bb-12 (MB), respectivamente. Na avaliação da resistência térmica e simulação gastrointestinal as micropartículas de Lactobacillus acidophillus La-5 (ML) apresentaram maior sobrevivência que Bifidobacterium Bb-12 (MB) nestas condições. Na viabilidade ao armazenamento somente as micropartículas Lactobacillus acidophillus La-5 (ML) mantiveram-se viáveis em todas as temperaturas avaliadas durante os 120 dias. Os tamanhos de partícula encontrados foram de 4.85 para Lactobacillus acidophillus La-5 (ML) e 8.75 para Bifidobacterium Bb-12 (MB), estando de acordo aos valores desejáveis para produtos obtidos por spray dryer. Por fim, as micropartículas de Lactobacillus acidophilus La-5 (ML) demostraram ser mais resistentes frente as condições avaliadas neste estudo.

ABSTRACT Polymeric stabilizers have received attention in the preparation of nanostructured systems due to their ability to enhance formulation stability. Considering this, the objective of this work was to prepare poly(ε-caprolactone) nanocapsules using the pullulan as a polymeric stabilizer. The nanocapsules were prepared using the interfacial deposition method of preformed polymers and they were characterized in terms of pH, average diameter, polydispersity index, zeta potential, beclomethasone dipropionate content, encapsulation efficiency, photostability and drug release profiles. The formulations showed physicochemical characteristics consistent with nanocarriers for drug delivery such as: average diameter lower than 270 nm, polydispersity indexes lower than 0.2, negative zeta potential (-22.7 to -26.3 mV) and encapsulation efficiencies close to 100%. In addition, the nanocapsules were able to delay the beclomethasone dipropionate photodegradation under UVC radiation and by the dialysis bag diffusion technique, the nanocapsules were able to prolong the drug release. Thus, pullulan could be considered an interesting excipient to formulate polymeric nanocapsules.

ABSTRACT

abstract Daptomycin (DPT) was the first lipopeptide antibiotic available for commercialization. It is active against gram-positive bacteria, including resistant strains. This work aimed to develop and validate a turbidimetric microbiologic assay to determine daptomycin in an injectable form. A 3x3 design was employed, at concentrations of 1, 2 and 4.0 µg/mL. The microorganism test used was Staphylococcus aureus ATCC 6538p, and Antibiotic Medium 3 was used as the culture medium. Method validation demonstrated that the bioassay was linear (r=0.9995), precise (RSD=2.58%), accurate (recovery 100.48± 2.11%), and robust. Degradation kinetics was also performed in an alkaline medium, indicating that daptomycin degradation follows first order kinetics under these conditions. The analyses of degraded solutions showed that daptomycin degradation products do not possess bactericidal activity. The bioassay was compared to HPLC method that was previously developed and no significant difference was found between them (p>0.05). The method proved to be appropriate for daptomycin injection quality control.


resumo A daptomicina (DPT) é o primeiro lipopeptídeo cíclico disponível para comercialização. Possui atividade frente a bactérias gram-positivas, incluindo cepas resistentes. O objetivo deste trabalho foi desenvolver e validar um ensaio microbiológico turbidimétrico para quantificar a daptomicina na forma injetável. Empregou-se delineamento 3x3, nas concentrações de 1,0; 2,0 e 4,0 µg/mL. Como micro-organismo teste foi usado Staphylococcus aureus ATCC 6538p, e Meio para Antibióticos nº 3 foi empregado como meio de cultura. A validação do método demonstrou que o ensaio foi linear (r=0,9995), preciso (RSD=2,55%), exato (recuperação de 100,48 ± 2,11%) e robusto. A cinética de degradação em meio alcalino foi avaliada, indicando que a daptomicina segue cinética de primeira ordem nessa condição. A análise das soluções degradadas mostrou que os produtos de degradação da daptomicina não possuem atividade antimicrobiana. O bioensaio foi comparado com o método por CLAE previamente desenvolvido e não houve diferença significativa entre ambos (p<0,05). O método mostrou-se apropriado para o controle de qualidade da daptomicina injetável.

ABSTRACT

O objetivo deste trabalho foi avaliar microcápsulas contendo Bifidobacterium animalis e Lactobacillus acidophilus, produzidas por spray drying. Ensaios de sobrevivência foram conduzidos para avaliar a resistência dos probióticos a condições gastrointestinais simuladas e a sua viabilidade durante 120 dias de armazenamento a 4ºC e 25ºC, além da análise morfológica das microcápsulas. A microencapsulação protegeu os probióticos das condições gastrointestinais simuladas, os quais permaneceram viáveis após 120 dias de armazenamento a 4ºC e 25ºC, sendo mais viáveis a 4ºC. As microcápsulas apresentaram forma esférica, com superfície contínua relativamente lisa e sem fissuras. O estudo indica que microcápsulas de B. animalis e L. acidophilus, produzidas por spray drying, sobrevivem a condições gastrointestinais simuladas e podem ser melhor armazenadas por 120 dias a 4ºC.


The aim of this study was to evaluate microcapsules containing Lactobacillus acidophilus and Bifidobacterium animalis by spray drying. Survival assays were conducted to evaluate the resistance of probiotics to simulated gastrointestinal conditions and its availability during 120 days of storage at 4°C and 25°C, besides morphological analysis of the microcapsules. Microencapsulation protected the probiotics from simulated gastrointestinal conditions, which also remained viable after 120 days of storage at 4ºC and 25ºC, but more viable at 4ºC. The microcapsules showed spherical shape with relatively smooth continuous surface without cracks. The study indicates that microcapsules of B. animalis or L. acidophilus by spray drying survive in simulated gastrointestinal conditions and can be better stored for 120 days at 4°C.

ABSTRACT

Microencapsulation is a process in which active substances are coated by extremely small capsules. It is a new technology that has been used in the cosmetics industry as well as in the pharmaceutical, agrochemical and food industries, being used in flavors, acids, oils, vitamins, microorganisms, among others. The success of this technology is due to the correct choice of the wall material, the core release form and the encapsulation method. Therefore, in this review, some relevant microencapsulation aspects, such as the capsule, wall material, core release forms, encapsulation methods and their use in food technology will be briefly discussed.


A microencapsulação é um processo em que substâncias ativas são revestidas por cápsulas extremamente pequenas. É uma tecnologia nova, a qual tem sido empregada na indústria de cosméticos, farmacêutica, agrotóxicos e alimentícia e, nesta, é utilizada em aromas, ácidos, óleos, vitaminas, micro-organismos, entre outros. O êxito nessa tecnologia deve-se à correta escolha do material encapsulante, da forma de liberação do núcleo e do método de encapsulação. Dessa forma, nesta revisão, serão abordados, sucintamente, alguns aspectos relevantes da microencapsulação, como a cápsula, o material encapsulante, as formas de liberação do núcleo, os métodos de encapsulação, assim como sua utilização na tecnologia de alimentos.

ABSTRACT

Superficial mycoses of the skin are among the most common dermatological infections, and causative organisms include dermatophytic, yeasts, and non-dermatophytic filamentous fungi. The treatment is limited, for many reasons, and new drugs are necessary. Numerous essential oils have been tested for both in vitro and in vivo antifungal activity and some pose much potential as antifungal agents. By using disk diffusion assay, we evaluated the antifungal activity of lemongrass oil and citral against yeasts of Candida species (Candida albicans, C. glabrata, C. krusei, C. parapsilosis and C. tropicalis). This study showed that lemongrass oil and citral have a potent in vitro activity against Candida spp.
